Among the misregulated genes in Ldb1−/− Flk1+ BL-CFCs directly bound by Ldb1 (Set1*), we found genes previously shown to participate in the regulation of mouse embryonic development and hematopoiesis such as Gata2,20,21 Gata1,22 Runx1,23 Fli1,24 Lyl1,25 Id2, Id4,26 Klf4,27 Gfi1b,28 Cbfa2t3/Eto2,29 Myb,30 Dkk1, Sfrp1,31,32 Meis1,33 Raldh2,34 and Fgfr2.35 The changes observed in some of those essential genes were confirmed by real-time polymerase chain reaction (supplemental Figure 3).

Several of these genes have a documented role in hematopoiesis, including Erg,20 Cited2,21 Ctnnb1,22 Fli1,23 Gata2,24 Id2,25 Id4,26 Ikzf1,27 Lyl1,28 Meis1,29 Pbx1,30,31 Runx1,32,33 Stat5,34 Tcf7,35 and Pml-RARA.36 Direct regulation of multiple hematopoiesis TFs by HoxB4 strongly implies that it is a master regulator positioned high in the hierarchy of the hematopoiesis-regulatory network.

Other potential factors regulating hemangioblast differentiation include differentiation-promoting transcription factors from the homeobox gene family [13] or differentiation-inhibiting transcription factors from the Id family [14]. Angioblasts have the ability to proliferate, migrate, and differentiate into endothelial cells, but have not yet acquired mature endothelial markers such as CD31 or von Willebrand factor (vWF).

It was reported recently that mice deficient in Id-2 lack lymph nodes and Peyer's patches, because they are deficient in a cell population essential for the generation of peripheral lymphoid organs (44).
